Long-term toxicity test
长期毒性实验又被称为反复给药毒性实验,研究动物连续给予受试物后对机体产生的毒性反应,目的是通过重复给药的动物实验表征受试物的毒性作用,降低临床受试者药物风险。
安全性评价
急性毒性实验;长期毒性实验;局部毒性实验:溶血、过敏、刺激性试验;遗传毒性实验:染色体畸变试验、微核试验、Ames试验;生殖发育毒性实验 ;致癌实验
急性毒性实验
急性毒性实验又称单次给药急性毒性实验,是指24h内一次或多次给予动物受试物后所产生的的毒性反应。 “一次”指经口、经注射途径染毒,经呼吸道和皮途径染毒时在规定期间内持续接触染毒的过程。
局部毒性实验
局部毒性实验主要是指化学药物经眼、耳、口、鼻、呼吸道、关节腔、皮肤、肌肉、粘膜组织等非口服途径给药的局部用药所产生的刺激性、过敏性、和溶血性实验。以提示临床应用时可能出现的毒性反应,保障临床用药的安全有效。
生殖发育毒性实验
三段生殖毒性试验是由美国食品药品管理局(FDA)提出的生殖毒性试验方案。主要用于评价药品的生殖发育毒性。
遗传毒性实验
Ames实验 Ames试验也叫做鼠伤寒沙门氏菌回复突变试验,是遗传毒理学研究的重要内容,可以检测药物是否具有致突变、致畸变和致癌变作用,是目前检测基因突变常用的方法之一,可进行药物的遗传毒性评价。
致癌实验
啮齿动物长期致癌实验是迄今为止最具说服力的致癌性评价模型,也是判断其它模型或方法敏感性和特异性的金标准。啮齿动物以适当染毒途径给予不同剂量受试物,观察动物最终发生肿瘤的种类和数量。
